The Positive Allosteric Modulation of alpha7-Nicotinic Cholinergic Receptors by GAT107 Increases Bacterial Lung Clearance in Hyperoxic Mice by Decreasing Oxidative Stress in Macrophages

Alex G. Gauthier et al.

Summary: This study suggests that a novel compound, GAT107, can improve lung balance and decrease bacterial burden by activating alpha 7 nicotinic acetylcholine receptor. GAT107 also rescues compromised phagocytic functions and attenuated oxidative stress in macrophages exposed to hyperoxia. Moreover, GAT107 increases the activity of superoxide dismutase 1, Nrf2 activation, and heme oxygenase-1 expression, indicating its potential use in improving host defense functions in patients exposed to prolonged periods of hyperoxia.
