Hereditary α tryptasemia is a valid genetic biomarker for severe mediator-related symptoms in mastocytosis

Georg Greiner et al.

Summary: The study showed that in patients with mastocytosis, those with HαT had higher levels of tryptase and were more likely to experience hymenoptera venom hypersensitivity reactions and severe cardiovascular mediator-related symptoms/anaphylaxis. This suggests that HαT could be a novel emerging robust biomarker in mastocytosis.

Phenotype and risk factors of venom-induced anaphylaxis: A case-control study of the European Anaphylaxis Registry

Wojciech Francuzik et al.

Summary: The study evaluated the phenotype and risk factors of venom-induced anaphylaxis (VIA) using data from the European Anaphylaxis Registry. VIA more frequently involved multiple organ systems and was associated with cardiovascular symptoms, while the absence of skin symptoms during anaphylaxis was correlated with a higher risk of severe reactions. Patients with intermediate baseline serum tryptase levels and without skin involvement had a higher risk of severe VIA. Patients receiving beta-blockers or angiotensin-converting enzyme inhibitors had a higher risk of developing severe cardiovascular symptoms in VIA and non-VIA cases, where patients experiencing VIA received epinephrine less frequently than cases with non-VIA.

Heritable risk for severe anaphylaxis associated with increased α-tryptase-encoding germline copy number at TPSAB1

Jonathan J. Lyons et al.

Summary: This study revealed that genotypes have a significant impact on the severity of anaphylaxis in humans, particularly in association with H alpha T. The prevalence of H alpha T was higher in healthy individuals and controls with nonatopic disease, and it was particularly prominent in systemic mastocytosis and idiopathic anaphylaxis, suggesting it as a potential risk factor for severe allergic reactions.

Patients with mast cell activation symptoms and elevated baseline serum tryptase level have unique bone marrow morphology

Matthew P. Giannetti et al.

Summary: Patients with MCAS-T exhibit unique morphologic and histologic features in bone marrow, including larger, hypogranular MCs located in paratrabecular and perivascular areas, associated with bone marrow eosinophilia. All patients available for tryptase genotyping were confirmed to have HaT, indicating that elevated tryptase levels in MCAS-T patients are caused by this genetic trait.
