Current and Emerging Clinical Treatment in Mitochondrial Disease

Rory J. Tinker et al.

Summary: Primary mitochondrial disease (PMD) is a group of complex genetic disorders caused by pathogenic variants in nuclear or mitochondrial genomes. Current treatment options are mostly supportive and preventive, with new pharmacological and non-pharmacological therapies undergoing clinical trials. Emerging therapies include novel pharmacological treatment, dietary supplementation, exercise training, device use, mitochondrial donation, tissue replacement gene therapy, hypoxic therapy, and mitochondrial base editing.

The Power of Yeast in Modelling Human Nuclear Mutations Associated with Mitochondrial Diseases

Camilla Ceccatelli Berti et al.

Summary: The yeast Saccharomyces cerevisiae is an excellent model system for studying mitochondrial diseases, offering advantages in validating and characterizing new mutations, dissecting gene functionality related to mitochondria, and discovering potential therapeutic molecules. Studies on this simple eukaryotic organism have contributed significantly to the understanding of mitochondrial diseases.

Bi-allelic KARS1 pathogenic variants affecting functions of cytosolic and mitochondrial isoforms are associated with a progressive and multisystem disease

Gerarda Cappuccio et al.

Summary: KARS1 mutations can cause severe neurological symptoms, including congenital and progressive microcephaly, seizures, developmental delay/intellectual disability, and cerebral atrophy. Additionally, oculo-motor dysfunction and immuno-hematological problems are common. Studies in yeast mutants suggest that lysine supplementation may have therapeutic potential in some patients.

LBSL Case Series and DARS2 Variant Analysis in Early Severe Forms With Unexpected Presentations

Menno D. Stellingwerff et al.

Summary: The study identified two distinct MRI phenotypes of LBSL cases, including early severe cerebral hypoplasia/atrophy and white matter abnormalities, with different levels of severity based on patient characteristics. The findings have significant implications for the diagnosis and understanding of LBSL disease mechanisms, suggesting a dominant neuronal/axonal involvement in severe cases.
