Summary: Subgenomic RNAs are essential for viral growth and pathogenicity, and within the flavivirus genus, two classes of Xrn1-resistant RNA motifs have similar structural configurations. Mutational analysis on tick-borne and no-known-vector flaviviruses showed that mutations targeting nucleotides involved in the structure features of Xrn1-resistant RNA motifs can affect the generation of subgenomic flaviviral RNA in vivo.

By studying xrRNA from Tamana bat virus (TABV), researchers discovered that different sequences and interactions can achieve the same overall architecture, providing insight into the diversity of this type of RNA and informing searches for undiscovered xrRNAs in viruses and beyond.
