Tracking break-induced replication shows that it stalls at roadblocks

Liping Liu et al.

Summary: Break-induced replication (BIR) repairs DNA double-strand breaks and plays a role in genome instability, with enzymes required for BIR synthesis identified. New research shows BIR synthesis starts soon after strand invasion, progresses slower than S-phase replication, and requires primase for stabilization of the nascent leading strand. DNA synthesis can initiate in the absence of certain enzymes, but efficiency is compromised. Interstitial telomeric DNA and transcription can disrupt and suppress BIR initiation, leading to mutagenesis and chromosome rearrangements.

Alternative paths to telomere elongation

Jennifer J. Lee et al.

Summary: Overcoming cellular senescence induced by telomere shortening is crucial in tumorigenesis, with most cancers maintaining telomeres through telomerase expression, while a subset use ALT for telomere elongation. The BIR mechanism, independent of RAD51, underlies ALT telomere synthesis, while homologous recombination dependent on RAD51 is necessary for ALT cancer cell maintenance. Despite evidence suggesting replication stress induces BIR at telomeres, ALT research is still in its early stages with much still unknown.

Beyond Kinases: Targeting Replication Stress Proteins in Cancer Therapy

Katherine E. Baillie et al.

Summary: DNA replication stress is a common state in many cancers, and targeting replication stress response factors such as ATR and CHK1 kinases is a promising therapeutic approach. However, research revealing the complexity of the replication stress response suggests new genetic interactions and candidate therapeutic targets. Many of these candidates regulate DNA transactions around reversed replication forks, including helicases, nucleases, and alternative polymerases that promote fork stability and restart.
