A toolkit based on an optimized zCas9i gene allows targeting multiple independent loci simultaneously. Efficient multiplexing was achieved in Nicotiana benthamiana and Arabidopsis thaliana, with the latter showing lower efficiency due to Cas9 availability limitations. This study opens new possibilities for generating complex genotypes through multiplexing.
