Donor-specific phenotypic variation in hiPSC cardiomyocyte-derived exosomes impacts endothelial cell function

Amy Turner et al.

Summary: The study found significant differences in RNA and miRNA content in exosomes derived from hiPSC-CMs of LVH-affected individuals compared to those from unaffected individuals, and these differences functionally affect the phenotypes of endothelial cells in a donor-specific manner. These insights provide valuable information for regenerative medicine and the use of stem cells and exosomes in cardiovascular therapeutic applications.

Interactome Analysis of iPSC Secretome and Its Effect on Macrophages In Vitro

Luca Tamo et al.

Summary: Macrophages play vital role in tissue repair and regeneration, and iPSC-CM has shown potential in reducing lung fibrosis and regulating macrophage phenotype. Through gene network analysis, this study identified AAP and ELAVL-1 in iPSC-CM as key components in regulating macrophage phenotype. Experimental confirmation demonstrated that AAP and ELAVL1 can change macrophage phenotype and reduce profibrotic characteristics, highlighting the antifibrotic and regenerative potential of iPSC-CM.

Current Status of Angiogenic Cell Therapy and Related Strategies Applied in Critical Limb Ischemia

Lucia Beltran-Camacho et al.

Summary: Critical limb ischemia is the most severe form of peripheral arterial disease, commonly caused by atherosclerosis leading to progressive blockage of arterial vessels. For patients not suitable for surgical intervention, angiogenic cell therapy has emerged as a promising alternative, showing potential for enhancing revascularization by promoting vessel formation and blood flow recovery.

Human heart-forming organoids recapitulate early heart and foregut development

Lika Drakhlis et al.

Summary: This study successfully generated three-dimensional heart-forming organoids that closely resemble early native heart anlagen, containing myocardial layer, endocardial-like cells, septum-transversum-like anlagen, and distinct anterior versus posterior foregut endoderm tissues. These organoids were used to study genetic defects in vitro and demonstrated a phenotype similar to cardiac malformations seen in transgenic mice with the knockout of NKX2.5 gene.
