dUTPase inhibition confers susceptibility to a thymidylate synthase inhibitor in DNA-repair-defective human cancer cells

Summary: Deficiency in DNA repair proteins increases susceptibility to DNA damage, making cancer cells more vulnerable to various cancer chemotherapies. 5-Fluorouracil inhibits thymidylate synthase and causes DNA damage, but the role of DNA repair pathways in its antitumor activity is still unclear. Inhibition of dUTPase leads to misincorporation of 5-FU and uracil into DNA, with base excision repair and homologous recombination pathways involved in repairing the resulting DNA damage.

Structural insights into an MsmUdgX mutant capable of both crosslinking and uracil excision capability

Summary: In this study, the high-resolution crystal structures of MsmUdgX H109E in free and DNA-bound forms were solved, revealing that it employs a similar excision-inhibition mechanism to the WT enzyme. The residues Arg107 and Gln53 were found to play important roles in the biochemical properties of MsmUdgX. These findings provide new insights into the catalytic process of MsmUdgX and enhance our understanding of this unique enzyme.
