Lipid Remodeling in Hepatocyte Proliferation and Hepatocellular Carcinoma

Summary: Our study demonstrates that specific lipid metabolic pathways are coherently altered when hepatocytes switch to proliferation, providing potential targets for therapeutic strategies and prognostic biomarkers of HCC. By integrating metabolomics, lipidomics, and transcriptomics, we have linked changes in the lipidome of proliferating hepatocytes to altered metabolic pathways including lipogenesis and generation of phosphatidylcholine. Additionally, we confirmed this altered lipid signature in human HCC and found a positive correlation of monounsaturated phosphatidylcholine with hallmarks of cell proliferation and hepatic carcinogenesis.

Metabolomic profile of cancer stem cell-derived exosomes from patients with malignant melanoma

Summary: Malignant melanoma, the most aggressive form of skin cancer, is characterized by its extraordinary metastasis capacity and chemotherapy resistance, mainly due to melanoma cancer stem cells. Current clinical biomarkers for this neoplasia are lacking, highlighting the urgent need for new biomarkers to enable early diagnosis and effective disease monitoring. Exosomes, a novel source of biomarkers, show significant metabolomic differences in MM CSC-derived exosomes and serum-derived exosomes from patients with MM compared to healthy controls, indicating their potential as clinically useful biomarkers for MM diagnosis.

Remodeling Lipids in the Transition from Chronic Liver Disease to Hepatocellular Carcinoma

Summary: Hepatocellular carcinoma (HCC) has a poor prognosis and is a leading cause of mortality worldwide. This study analyzed lipid alterations in blood and tissues of HCC patients compared to healthy controls and those with chronic liver disease. The research identified significant changes in lipid composition, particularly in triacylglycerides and phosphatidylcholines, between healthy controls, CLD patients, and HCC patients. These findings may have implications for the prevention, early detection, and treatment of HCC.

Mass spectrometry imaging of untreated wet cell membranes in solution using single-layer graphene

Summary: This study demonstrates a method for sputtering atomic and molecular secondary ions through single-layer graphene to enable SIMS imaging of untreated wet cell membranes at subcellular resolution. The findings show that the method allows observation of lipid distribution in cell membranes without labeling, and that cells remain intact during imaging. Additionally, molecular dynamics calculations and ion dose-dependence studies suggest that sputtering through graphene occurs through a transient hole in the graphene layer.

Lipidomic Signatures for Colorectal Cancer Diagnosis and Progression Using UPLC-QTOF-ESI+MS

Summary: Metabolomics combined with bioinformatics can identify relevant biomolecules and potential biomarkers related to specific metabolic pathways in colorectal cancer. This study utilized metabolomic profiling to identify over 400 metabolites in blood serum samples from CRC cases, confirming a specific lipid phenotype and upregulated lipogenesis associated with tumor progression. Both multivariate and univariate bioinformatics analyses supported the importance of putative lipid biomarkers in altered metabolic pathways of colorectal cancer.

Summary: Fatty acid metabolism is closely related to tumor occurrence and development, with specific fatty acids influencing the activity and migration of tumor cells. Five specific fatty acids may serve as potential tumor markers in invasive ductal carcinoma (IDC), and the expression level of fatty acid synthase (FASN) affects the serum fatty acid profiles of IDC patients. Additionally, FASN-mediated changes in specific fatty acids promote tumor cell migration.
