Oxidative Stress, GTPCH1, and Endothelial Nitric Oxide Synthase Uncoupling in Hypertension

Yin Wu et al.

Summary: This review highlights the importance of nitric oxide in maintaining vascular homeostasis and its role in hypertension. Recent studies suggest that BH4 treatment could be a promising therapeutic for hypertension, but abnormalities in BH4 availability can trigger pathological events that affect blood pressure regulation. Continued research in this area is crucial to improve potential therapies for treating hypertension.

Nrf2 inhibition induces oxidative stress, renal inflammation and hypertension in mice

Zeba Farooqui et al.

Summary: In this study, mice treated with Nrf2 inhibitor ML385 and pro-oxidant BSO exhibited exacerbated hypertension, increased oxidative stress, and renal dysfunction. Compared to mice treated with only BSO, co-treatment with ML385 and BSO showed increased activity of phase II antioxidant enzyme and significant alterations in inflammatory cytokine levels.

Targeting feed-forward signaling of TGF beta/NOX4/DHFR/eNOS uncoupling/TGF beta axis with anti-TGF beta and folic acid attenuates formation of aortic aneurysms: Novel mechanisms and therapeutics

Kai Huang et al.

Summary: This study identified novel mechanisms and potential treatments for thoracic aortic aneurysm (TAA) in Fbn1(C10)(39G)(/+) Marfan Syndrome (MFS) mice, highlighting the central role of uncoupled eNOS in TAA formation. Anti-TGF beta and folic acid diet were shown to effectively abolish aneurysm formation through inactivation of a novel TGF beta/NOX4/DHFR/eNOS uncoupling/TGF beta feed-forward pathway.

Catalytic Antioxidants in the Kidney

Yu Ah Hong et al.

Summary: Reactive oxygen and nitrogen species play crucial roles in kidney injuries, making antioxidant agents, especially catalytic antioxidants that mimic antioxidant enzymes, effective in combating oxidative stress-related kidney diseases. Research has shown the effectiveness of several catalytic antioxidants in treating disease models associated with oxidative stress.

Summary: NOXs are a family of proteins that produce reactive oxygen species, found in various cell and organ systems. The evolutionary conservation highlights their importance, with research focusing on their mechanisms of action and role in oxidative stress and disease, starting from their discovery in vasculature. Studies have also used isoform-selective inhibitors to investigate the role of NOX isozymes in vascular biology, particularly in inflammation, pulmonary hypertension, and aging.
