A modular master regulator landscape controls cancer transcriptional identity

Evan O. Paull et al.

Summary: Using a network-based approach, 407 master regulator proteins were identified to canalize individual cancer samples into 112 transcriptionally distinct tumor subtypes. These proteins were organized into 24 pan-cancer master regulator block modules regulating key cancer hallmarks and predictive of patient outcome. Genomic alterations were predicted to induce aberrant MR activity, providing insight into mechanisms linking tumor genetics and transcriptional identity.

Proteogenomic and metabolomic characterization of human glioblastoma

Liang-Bo Wang et al.

Summary: Understanding the molecular pathogenesis of glioblastoma is crucial for improving diagnosis and treatment. Integrated analysis of genomic, proteomic, and metabolomic data on treatment-naive glioblastomas has identified key phosphorylation events and potential targets for specific tumor alterations. Immune subtypes with distinct immune cell types were discovered, and specific histone acetylation patterns were correlated with these subtypes, providing insights for stratification of patients for more effective treatment.

Germline genetic contribution to the immune landscape of cancer

Rosalyn W. Sayaman et al.

Summary: Understanding the host's genetic background's contribution to cancer immunity has significant impact on the composition and functional orientation of the tumor immune microenvironment, with certain genes potentially acting as immune modulators. Investigating the effects of germline variants on immune traits provides a resource toward further understanding tumor-immune interactions.

A balance score between immune stimulatory and suppressive microenvironments identifies mediators of tumour immunity and predicts pan-cancer survival

Tolga Turan et al.

Summary: The balance between immune-stimulatory and immune-suppressive mechanisms in the tumour microenvironment is crucial for predicting cancer outcomes and the efficacy of immune checkpoint-inhibition therapies. The MIRACLE score, which deconvolves T cell infiltration from inhibitory mechanisms, outperforms current immune signatures in predicting survival and response to checkpoint-inhibitor therapies. This highlights the importance of considering both activatory and suppressive factors in the immune response against cancer.
