An 18-gene signature based on glucose metabolism and DNA methylation improves prognostic prediction for urinary bladder cancer

Zhuonan Liu et al.

Summary: Glucose metabolism and DNA methylation are important in cancers. A prognostic signature for bladder cancer was established using 18 genes associated with glucose metabolism and DNA methylation. Two genes (UCHL1 and PYCR1) were shown to enhance the malignant characteristics of bladder cancer cells.

Gene co-expression network analysis reveals immune cell infiltration as a favorable prognostic marker in non-uterine leiomyosarcoma

Mohammad Darzi et al.

Summary: This study aimed to explore prognostic genes of non-uterine leiomyosarcoma (NULMS) through system biology approaches, identifying seven co-expression modules and immune-related pathways. The study suggests NULMS as a good candidate for immunotherapy and potential genes for targeted therapy.

Targeting the Proline-Glutamine-Asparagine-Arginine Metabolic Axis in Amino Acid Starvation Cancer Therapy

Macus Tien Kuo et al.

Summary: Proline, glutamine, asparagine, and arginine are conditionally non-essential amino acids that are essential for the growth of highly proliferative cells such as cancers. Targeting these amino acids through amino acid starvation strategies has shown promise in clinical applications, but drug resistance remains a major obstacle, primarily due to reactivation of key enzymes and reprogramming of compensatory survival pathways.

Reprogramming of mitochondrial proline metabolism promotes liver tumorigenesis

Zhaobing Ding et al.

Summary: Dysregulated cellular energetics is a recognized hallmark of cancer, with metabolic reprogramming playing a crucial role in sustaining cancer cell proliferation. The proline metabolism pathway is implicated in promoting tumorigenesis in various cancer types, including liver cancer. Research on the impact of proline metabolism on liver cancer and other cancers, as well as the related mechanisms, is a priority in current therapeutic studies.

Summary: PYCR is an important enzyme for L-proline biosynthesis, with mutations in the PYCR2 gene linked to postnatal microcephaly and hypomyelination disorders. The disease-related variants R119C and R251C show catalytic impairment and structural defects, contributing to the pathology of these disorders.

MiR-328-3p inhibits lung adenocarcinoma-genesis by downregulation PYCR1

Jiancong Lu et al.

Summary: In NSCLC, the majority of patients have lung adenocarcinoma (LA) with varying survival rates. PYCR1 gene and miR-328-3p are associated with cancer development, with miR-328-3p targeting PYCR1 to suppress LA cell malignancy by inhibiting cell proliferation and migration while promoting apoptosis.

The metabolism of cancer cells during metastasis

Gabriele Bergers et al.

Summary: This review discusses the metabolic rewiring in cancer cells during the metastatic cascade and the distinct metabolic features of primary tumors and metastases. Metastasis is the leading cause of death in most cancer patients, and targeting metastatic seeding and colonization remains a significant challenge. Adjusting metabolism may provide therapeutic opportunities to prevent or reduce metastasis, and targeting the new metabolic traits in established metastases could lead to their eradication.
