Circadian/multidien Molecular Oscillations and Rhythmicity of Epilepsy (MORE)

Summary: Seizures in individuals with epilepsy tend to occur more frequently at specific times, exhibiting a circadian rhythmicity. The MORE hypothesis suggests that molecular oscillations of gene and protein expression may contribute to the timing of seizure genesis. Understanding these molecular oscillations could lead to new therapeutic targets and predictive biomarkers for epilepsy.

Summary: Research using a multi-omics approach in an animal model of mesial temporal lobe epilepsy revealed differences in the transcriptomic and proteomic profiles between the dentate gyrus (DG) and Cornu Ammonis 3 (CA3) of the hippocampus, suggesting enhanced epileptogenesis in the CA3 region and abnormalities mainly occurring in CA3. Abnormal regulation of excitatory neuronal mechanisms mediated by NMDA receptors, changes in serotonin signaling, and neuronal activity controlled by CaMK regulation and LRRK2/WNT signaling pathways are involved in the epileptogenesis in the pilocarpine model.

These differences are driven by intra-hippocampal and entorhinal inputs, supported by the interaction between excitatory gamma inputs and local inhibition.

Spatio-temporal heterogeneity in hippocampal metabolism in control and epilepsy conditions

Summary: The hippocampus in control mice shows spatial and time-dependent differences in energy metabolism along the dorsoventral axis, with changes in metabolic activity between morning and afternoon. In an epilepsy model, this spatiotemporal organization is significantly altered, with enhanced aerobic glycolysis in both dorsal and ventral parts in the afternoon when seizure probability is high. This suggests that energy metabolism is tailored to the functions performed by brain networks and alterations in pathological conditions may contribute to network dysfunctions.
