Luengo et al. (2020) demonstrate that overactivation of pyruvate dehydrogenase (PDH) impairs NAD(+) regeneration by altering mitochondrial membrane potential and driving ATP synthesis beyond demand, leading some cells to prioritize aerobic glycolysis to meet the requirement for oxidized cofactors in biosynthetic metabolism.
