Identification of highly active natural thyroid hormone receptor agonists by pharmacophore-based virtual screening

Summary: In this study, novel hit compounds with higher binding activity than crystal TR ligands were identified through in silico virtual screening, followed by investigation of their binding mode and action mechanism using molecular docking and molecular dynamics studies. Two compounds, UNPD19665 and UNPD184785, were found to have binding activity values of 9.82 nM and 12.62 nM, respectively, outperforming the crystal TR ligands.

Summary: In this study, various ROR gamma t agonists were investigated using multiple molecular modeling techniques to understand the structural requirements for designing better active compounds. The CoMFA and CoMSIA models showed satisfactory predictive ability, while molecular docking and dynamics simulations identified key residues for higher binding activity in the active site of ROR gamma t.
