Summary: This study investigated the effect of mutations on PSEN1 and PSEN2 genes on the BBB function using induced pluripotent stem cells (iPSCs). The results showed that iPSC-derived BMECs from FAD patients with a mutation in the PSEN1 gene exhibited impaired barrier function, poor tight junction complexes, reduced drug efflux pump activity, reduced bioenergetics, and elevated A beta 1-40 peptides secretion, suggesting a more detrimental phenotype compared to PSEN2 mutation.
