Whole-exome Sequencing Reveals New Potential Susceptibility Genes for Japanese Familial Pancreatic Cancer

Summary: The study aimed to identify novel genes that increase the risk of familial pancreatic cancer (FPC) in the Japanese population. The findings revealed potentially harmful germline variants in tumor suppressor genes in FPC patients, and somatic loss of heterozygosity of certain FPC susceptibility genes may contribute to FPC development. Genetic testing for a wider range of FPC predisposition genes could improve screening methods for high-risk groups of pancreatic cancer.

Characterizing allele- and haplotype-specific copy numbers in single cells with CHISEL

Summary: Single-cell barcoding technologies enable simultaneous genome sequencing of thousands of individual cells, but with low sequencing coverage per cell. A method called CHISEL was introduced to infer allele- and haplotype-specific copy numbers in single cells and subpopulations by aggregating sparse signal across hundreds or thousands of individual cells. Application of CHISEL to single-cell sequencing datasets from breast cancer patients identified extensive allele-specific copy-number aberrations affecting genomic regions containing well-known breast cancer genes.

Copy neutral loss of heterozygosity (cnLOH) patterns in synchronous colorectal cancer

Summary: The study established a cnLOH profile of synchronous colorectal cancer (SCRC) and found associations between cnLOH and polyclonal SCRC, with inactivation of tumor suppressor genes and cnLOH being rare events in SCRC cases. The research defines distinct patterns of cnLOH in SCRC and provides initial evidence of a role for cnLOH in the etiology of SCRC.

HLA loss of heterozygosity-mediated discordant responses to immune checkpoint blockade in squamous cell lung cancer with renal metastasis

Summary: This study reported a case of squamous cell lung carcinoma with renal metastasis treated with pembrolizumab, showing different responses between the primary tumor and rare renal metastases. Loss of heterogeneity in HLA genes and high levels of intratumor heterogeneity were found to impact therapy resistance during tumor evolution, suggesting that they should also be considered during immunotherapy along with tumor mutational burden and PD-L1.

Overcoming resistance to endocrine therapy in hormone receptor-positive human epidermal growth factor receptor 2-negative (HR+/HER2-) advanced breast cancer: a meta-analysis and systemic review of randomized clinical trials

Summary: This meta-analysis and systematic review evaluated the efficacy of various targeted therapies in combination with endocrine therapy for HR+/HER2(-) advanced breast cancer. Addition of cyclin-dependent kinase 4/6 inhibitors to endocrine therapy significantly improved progression-free survival, overall survival, and tumor response rates with manageable toxicities. Mammalian targets of rapamycin inhibitors and exemestane did not show clinical benefit, while combination of phosphatidylinositol-4,5-bisphosphate 3-kinase inhibitors with fulvestrant demonstrated moderate improvement in progression-free survival but notable toxicities. Future studies are needed to optimize patient selection and dosing regimens for these available options.
