Laboratory evolution of a sortase enzyme that modifies amyloid-β protein

Summary: This study described the laboratory evolution of the bacterial transpeptidase sortase A to recognize specific sequences in the endogenous amyloid-beta (Aβ) protein, enabling site-specific protein modification without genetic manipulation. The evolved sortase showed a significantly changed substrate preference and was able to label endogenous Aβ in human cerebrospinal fluid with high sensitivity, demonstrating potential future applications in sortase-mediated labeling of Aβ peptides.
