Investigating the pernicious effects of heparan sulfate in serum amyloid A1 protein aggregation: a structural bioinformatics approach

G. Chandrasekhar et al.

Summary: Amyloid-A mediated (AA) amyloidosis is a pathogenic condition resulting from prolonged exposure to inflammatory conditions, causing aggregation of mutated/misfolded SAA1 protein in various tissues and organs. Recent studies suggest that genetic polymorphism G90D may cause enhanced association of SAA1 protein with heparan sulphate (HS), potentially exacerbating amyloidosis. This could lead to proteopathic incendiary behind AA amyloidosis and should be considered in future therapeutic development.

Binding mechanism and structural insights into the identified protein target of COVID-19 and importin-alpha with in-vitro effective drug ivermectin

Parth Sarthi Sen Gupta et al.

Summary: Ivermectin shows strong affinity to the RNA dependent RNA polymerase (RdRp) of COVID-19, with better structural stability compared to its interaction with the Helicase NCB site and Importin-alpha. Further experimental exploration and drug design could lead to more effective treatments for COVID-19.

FDA-approved thiol-reacting drugs that potentially bind into the SARS-CoV-2 main protease, essential for viral replication

Naun Lobo-Galo et al.

Summary: Given the current lack of specific antiviral treatment or vaccine for COVID-19, the search for drugs that can potentially inhibit essential viral enzymes is urgent. Through in silico testing, disulfiram and its metabolites are identified as potential antiviral drugs, with a proposed mechanism for their action.

Identification of new anti-nCoV drug chemical compounds from Indian spices exploiting SARS-CoV-2 main protease as target

Umesh et al.

Summary: Chemical compounds from Indian spices, such as Carnosol, Arjunglucoside-I, and Rosmanol, have shown potential as inhibitors of SARS-CoV-2 main protease in molecular docking experiments. These compounds exhibit strong and stable binding affinity with the active site of SARS-CoV-2 Mpro, suggesting they could be promising candidates for anti-viral drugs against nCoV pending further validation through clinical trials.

In-silico homology assisted identification of inhibitor of RNA binding against 2019-nCoV N-protein (N terminal domain)

Phulen Sarma et al.

Summary: This study targeted the RNA-binding N terminal domain of the N protein of coronavirus to identify possible inhibitors of RNA binding. Theophylline and pyrimidone derivatives were identified as potential inhibitors, opening up new avenues for in vitro validations of their RNA binding inhibition capabilities.

Summary: Through bioinformatics and computational modeling, ninety seven antiviral secondary metabolites from fungi were found to have high binding affinities on five SARS-CoV2 enzymes. Among them, Quinadoline B (19) showed favorable ADMET values and high gastrointestinal absorptive probability but poor blood-brain barrier crossing capacities.
