Phosphorylation in two discrete tau domains regulates a stepwise process leading to postsynaptic dysfunction

Peter J. Teravskis et al.

Summary: Mislocalization of Tau protein to dendritic spines and associated postsynaptic deficits are mediated through distinct phosphorylation sites. Phosphorylation at different domains of Tau protein leads to sequential processes resulting in synaptic dysfunction. Glycogen synthetase kinase 3 beta and cyclin-dependent kinase 5 play crucial roles in preventing Tau mislocalization to dendritic spines.

The β-Secretase BACE1 in Alzheimer's Disease

Harald Hampel et al.

Summary: BACE1 is a well-validated therapeutic target for slowing down Aβ production in early AD, despite previous clinical trials being discontinued. Further research should focus on the roles of BACE1 and Aβ in physiological and pathophysiological processes, as well as the substrates and downstream effects of BACE1 and BACE2.
