Digital holographic tomography (DHT) can measure the 3D refractive-index map of a microscopic specimen. To obtain the 3D map using a 2D camera, existing methods scan the angle of illumination, rotate the sample, scan the objective-lens focus, or change the wavelength. Here, we present a snapshot DHT technique that does not rely on a scanning mechanism. Using a technique named snapshot holographic optical tomography (SHOT), we acquire the 3D refractive-index maps of a polystyrene bead and a single Henrietta Lacks (HeLa) cell.
