In CD4+ T cells, HIV-1 buds from the host cell plasma membrane. The viral Gag polyprotein is mainly responsible for this process. However, the intimate interaction of Gag and lipids at the plasma membrane as well as its consequences, in terms of lipids lateral organization and virus assembly, is still under debate. In this review we propose to revisit the role of plasma membrane lipids in HIV-1 Gag targeting and assembly, at the light of lipid membranes biophysics and literature dealing with Gag-lipid interactions.
