The application of in-cell click chemistry to the visualization of chemical probes in chemical biology experiments is reviewed. Fluorometric dyes can perturb the distribution, and function, of a labelled chemical tool - probes incorporating 'silent' clickable handles can therefore report on more relevant subcellular distribution and biomolecular interaction events. The influence this research has had on target validation and molecular mode of action studies is also highlighted.
