A novel and efficient extraction technique is proposed in this paper. Dried fruiting bodies of Ganoderma lucidum (Lingzhi) were finely pulverized into ultrafine powder, subsequent to extract the antioxidant crude polysaccharides by ultrasonic circulating extraction. Several important extraction factors were investigated by single factor analysis using DPPH radical scavenging rate as an index. The extraction process was optimized by Uniform design using polysaccharide concentration as the index. Then the multiple regression equation was worked out, by which the optimal processing condition can be predicted, and the predicted model was experimentally verified. The highest polysaccharides concentration, i.e. 47.87 mg/ml, along with the highest DPPH center dot scavenging rate 53.63% can be achieved at 671 W ultrasonic power, 48 degrees C extraction temperature and 5.5/1 of intermit-running ratio (s/s), as well as 1:12.5 of solid-liquid ratio (w/v) and 45 min extraction time. The reproducible results confirmed that this novel technique was efficient for high extract yield of antioxidant crude polysaccharides from Lingzhi. (C) 2013 Taiwan Institute of Chemical Engineers.
