A maltogenic amylase from fenugreek (Trigonella foenum graecum) seeds was extracted and then purified to homogeneity. Optimization extraction was carried out using Box Benkhen Design (BBD). An overall purification of about 239-folds with an activity yield of 47% was achieved. The pure enzyme has an apparent molecular mass of 68 kDa, as determined by SDS-PAGE analysis. Its maximal activity was in the pH 5 and at 60 degrees C and the specific activity, under these conditions, was found to be 258 U/mg. This amylase showed an exo-specificity profile since it generated mainly maltose from starch. It exhibited a broad range of pH stability, lead to expect a great commercial value and a good prospect for industrial applications. (C) 2012 Elsevier B.V. All rights reserved.
