Microglia-mediated calcium-permeable AMPAR accumulation in the nucleus accumbens drives hyperlocomotion during cocaine withdrawal

Summary: In this study, the role of microglia in neuroadaptations in NAc synapses during cocaine withdrawal was investigated. Microglia depletion prevented cocaine-induced changes in dendritic spines and the accumulation of CP-AMPAR, and also prevented conditioned hyperlocomotion. These results suggest that microglia could be a promising therapeutic target for preventing relapse in substance abuse.

Gonadal hormones impart male-biased behavioral vulnerabilities to immune activation via microglial mitochondrial function

Summary: There is a strong male bias in many neurodevelopmental disorders such as autism spectrum disorder, but the mechanisms behind this bias are unknown. Infection during the perinatal period increases the risk of neurodevelopmental disorders. Using a mouse model, researchers found that early-life immune activation only induced deficits in social behaviors in male mice. These behavioral changes were associated with alterations in microglial immune signaling, mitochondrial morphology, gene expression, and function specifically in microglia, the brain's innate immune cells. Furthermore, the vulnerability to early-life immune activation and its effects on behavior and microglial mitochondria were programmed by the male-typical perinatal gonadal hormone surge. These findings highlight the role of microglia-specific mechanisms shaped by early developmental events in regulating male social behavior throughout lifespan.

Macrophages-derived exo-miR-4449 induced by Cryptococcus affects HUVEC permeability and promotes pyroptosis in BEAS-2B via the HIC1 pathway

Summary: Macrophages play a significant role in the progression of cryptococcosis. This study investigated the potential involvement of macrophage-derived exosomes in the pathogenesis of cryptococcosis. The study analyzed the changes of microRNAs in macrophage exosomes and explored the effects of highly expressed exo-miRNAs on human endothelial cell permeability, ROS accumulation, and pyroptosis in bronchial epithelioid cells. The results showed that exo-miR-4449 plays a crucial role in the pathogenesis of cryptococcosis and holds promise as a significant biomarker for treatment.

Neuronanomedicine for Alzheimer's and Parkinson's disease: Current progress and a guide to improve clinical translation

Summary: Neuronanomedicine is a multidisciplinary field that aims to develop innovative nanotechnologies for the treatment of neurodegenerative disorders. Nanoparticles are an important component of neuronanomedicine, as they can improve drug properties and enhance safety and delivery across the blood-brain barrier. However, a major challenge for neuronanomedicine translation is the poor understanding of bio-nano interactions, partly due to inconsistent reporting in published works. This review provides specific recommendations to guide researchers in harnessing the unique properties of nanoparticles for breakthrough treatments of Alzheimer's and Parkinson's disease.

Summary: Recent studies have found a negative association between childhood trauma and cognitive functioning in patients with schizophrenia. This study aimed to test the hypothesis that physical neglect is associated with cognitive ability, and that this association is mediated by inflammatory response and moderated by genetic risk for schizophrenia. The results showed that inflammatory response mediated the association between physical neglect and cognitive functioning, and genetic risk for schizophrenia moderated the direct pathway between physical neglect and non-social cognitive functioning.

Low-dose ionizing radiation promotes motor recovery and brain rewiring by resolving inflammatory response after brain injury and stroke

Summary: This study found that low-dose ionizing radiation (LDIR) reduces brain lesion size and reverses motor deficits in traumatic brain injury (TBI) and stroke. Transcriptomic analysis revealed LDIR-induced upregulation of anti-inflammatory and phagocytosis-related genes, and downregulation of pro-inflammatory cytokines. Live-cell assays confirmed the increased chemotactic and phagocytic capacities of microglia after LDIR. Furthermore, LDIR promoted axonal projections and brain activity recovery months after stroke. These findings suggest that LDIR could be a promising therapeutic strategy for TBI and stroke.
