Summary: This study investigated the expressions of miR-361-3p and KMT2A in AML tissues and cell lines, and identified an advanced target for AML therapy. The results showed low expression of miR-361-3p and high expression of KMT2A. Inhibition of KMT2A prevented AML cells from proliferating, migrating, and invading. KMT2A was also confirmed as a direct target of miR-361-3p and negatively correlated with it.

Summary: This study evaluates the consistency of erythrocyte volume parameters between Sysmex-XN9000 and CellaVisionDI-60. The results show that both instruments have good accuracy and consistency in measuring heterogeneity degree. It is suggested to combine Sysmex-XN9000 and CellaVisionDI-60 for comprehensive anisocytosis evaluation in daily work.

Summary: This study aimed to explore the efficacy of prophylactic pegfilgrastim in preventing febrile neutropenia (FN) in pediatric/adolescent cancer patients. The results showed no significant difference in reducing the rate of FN between pegfilgrastim and conventional methods. However, in patients receiving pegfilgrastim, rates of FN, grade 4 FN, severe neutropenia, and treatment delays due to FN still existed.

Summary: Wilms' tumor gene 1 (WT1) plays a crucial role in the biological and pathological processes of several human malignancies, especially in hematological malignancies such as leukemia and myelodysplastic syndrome. WT1 overexpression or mutation is commonly found in acute myeloid leukemia patients, and it has been widely regarded as a marker for monitoring minimal residual disease. Many researchers are interested in developing targeted therapy for WT1. This review provides a summary of the functions, correlation with other genes, clinical features, prognosis value, and targeting therapy of WT1 in hematological malignancies.

Summary: This study aimed to establish local reference intervals (RIs) for complete blood cell (CBC) counts in newborns. Capillary blood specimens were collected from 497 healthy newborns, and the RIs for 18 CBC parameters were calculated following the guideline. The conformity rate of the RIs in the validation specimens was 90%-100%. Conclusion: Region-specific RIs for CBC parameters in healthy newborns were established to aid in the diagnosis of hematological diseases in neonates.

Summary: This study aimed to evaluate whether there is a shift in risk stratification at progression compared to that at diagnosis in patients with MM, and to analyze its prognostic impact. The results showed that re-staging and stage migration have predictive value for prognosis, and patients with deteriorated stage migration or maintained advanced stage at progression may require more individualized treatment.

Summary: The aim of this study was to identify novel prognostic and therapeutic targets associated with acute myeloid leukemia (AML). The researchers found that circPLXNB2 was highly expressed in AML patients and cells, and it regulated tumor progression through the circPLXNB2/miR-654-3p/CCND1 axis. Therefore, circPLXNB2 may be a new therapeutic target for AML treatment.

Summary: This article presents a family with the beta-hemoglobin variant Hb Santa Juana (HBB:c.326A>G, beta 108(G10) Asn>Ser), also known as Hb Serres, spanning three generations. All affected family members had an abnormal hemoglobin fraction as detected by HPLC, but had normal blood count without signs of anemia or hemolysis. Oxygen affinity (p(50) (O-2) = 31.9-40.4 mmHg) was decreased in all probands, compared to 24.9-28.1 mmHg in unaffected individuals. Clinical symptoms likely related to the hemoglobin variant included cyanosis during anesthesia, while other complaints such as shortness of breath or dizziness were less clearly linked with the hemoglobin variant.

Summary: This study explored the clinical features and outcomes of patients with multiple myeloma harboring 1q21+ gene abnormality. The results showed that 1q21+ was associated with higher ISS stage, elevated lactate dehydrogenase, lower levels of hemoglobin and platelets, and coexistence of del(13q). Patients with 1q21+ had shorter progression-free survival and overall survival, and 1q21+ was identified as an independent adverse prognostic factor.

Summary: This study evaluated the characteristics of patients with hematological malignancies and SARS-CoV-2 infection and analyzed the risk factors for severity and mortality. The study found that acute myeloid leukemia and primary hematological disease in active state were independent risk factors for severe SARS-CoV-2 infection, and patients with AML had a higher risk of non-survival compared to patients with other types of HM. Additionally, waiting 14 days for chemotherapy initiation after negative SARS-CoV-2 testing was unnecessary.

Summary: Adult patients with newly diagnosed de novo acute myeloid leukemia (AML) with poor prognosis, known as type 1 primary refractory (REF1), were retrospectively analyzed to evaluate the impact of different salvage regimens on their response and overall survival (OS). The study found no significant difference in outcome among traditional salvage regimens in REF1 patients and suggested that G-CSF primed less intensive chemotherapy could serve as an alternative to intensive chemotherapy, while allo-HSCT is essential for long-term survival.

Summary: Patients with hematologic malignancies are at a higher risk of severe COVID-19 due to their immunodeficiency. A retrospective study of 289 patients in Zhejiang Province showed that older age, comorbid conditions, and active antineoplastic therapy were associated with increased risk of severe COVID-19. Patients with severe disease had abnormal levels of lymphocytes and inflammatory markers, and a significant proportion experienced persistent symptoms. The effectiveness of inactivated vaccines in these patients was uncertain.
