Precise Distance Measurements in DNA G-Quadruplex Dimers and Sandwich Complexes by Pulsed Dipolar EPR Spectroscopy

Summary: DNA G-quadruplexes have a tendency to form higher-order structures, and square-planar Cu(pyridine)(4) complexes are used as rigid spin labels to detect dimeric structures and measure intermolecular distances via pulsed dipolar EPR spectroscopy. Different spatial dimensions of G-quadruplex dimers and new binding modes of well-known G-quadruplex binders were discovered in this study. The transition metal labeling approach combined with pulsed EPR spectroscopy opens new possibilities for examining structures of non-covalent DNA aggregates.

Summary: Trimethylsilyl (TMS) groups are excellent NMR probes for biological macromolecules, and a system for genetic encoding of N-6-(((trimethylsilyl)-methoxy)carbonyl)-l-lysine (TMSK) has been developed for site-specific incorporation into proteins. TMSK can be readily identified in high-molecular weight systems without isotopic labeling, providing an inexpensive and convenient tool for studying protein structure and function. The utility of TMSK has been demonstrated in detecting ligand binding, measuring conformational change rates, and assessing protein dimerization.

Origin of the Isomer Stability of Polymethylated DOTA Chelates Complexed with Ln3+ Ions

It was found that electrostatic and steric repulsions play key roles in destabilizing certain isomers, and potential energy barriers for inter-conversion via arm rotation were also calculated. The possibility of designing better chelates for Ln(3+) ions for molecular medicine was discussed.

A Chiral Lanthanide Tag for Stable and Rigid Attachment to Single Cysteine Residues in Proteins for NMR, EPR and Time-Resolved Luminescence Studies

Summary: The study introduces a new stable chiral Ln(III) tag, known as C12, for site-specific attachment to proteins, enabling protein probing through various spectroscopic techniques. The C12 tag exhibits enhanced reactivity towards selenocysteine, offering a pathway for selectively tagging selenocysteine in proteins containing cysteine residues. With Tb-III or Tm-III ions, the C12 tag can generate pseudocontact shifts in protein NMR spectra, providing a rigid tether between lanthanide and protein for interpretation of the shifts and distance measurements in double electron-electron resonance experiments. Additionally, the Tb-III complex upon reaction with cysteine or thiol compounds shows a significant enhancement in luminescence quantum yield, making it a sensitive luminescence probe for time-resolved FRET assays and enzyme reaction monitoring.
