This study was aimed at the applications of an ex vivo assays to characterization of CEM (Human T-Lymphoblastoid) cells CEM cells were cultured in three dimensional (3-D) geometry in the Hollow Fibre Bioreactor (HFB) device. The cells were treated with Herceptin, anti-HER-2 (clone CB-11) and lipoplex containing lipofectamine (LipA) and plasmid DNA. To identify the response to treatment, the viability was established using Trypan blue assays Magnetic resonance imaging (MRI) at 94 Tesla (T) was applied for localization of the cells in the HFB device The structural changes in the cells associated with treatment were examined with matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS). The tryptic peptides and glycopeptides detected in treated cells provided evidence of the efficacy of antibody binding to the receptor The results of the study confirmed that cells growth significantly decreased after treatment with antibodies and transfection with lipoplex Crown Copyright (C) 2010 Published by Elsevier Masson SAS.
