DNA replication is a fragile process, since unavoidable lesions in the template DNA cause replicative polymerases to stall, posing a serious threat to genome integrity. Homologous recombination, translesion DNA synthesis and de novo reinitiation of DNA synthesis ensure robust replication by navigating it passed damaged DNA. In this review, we highlight the relationship between these three processes. (c) 2006 Elsevier Inc. All rights reserved.
