Herein we report a simple and efficient one-pot three-component synthesis of 5-phosphonato-3,4-dihydropyrimidin-2(1H)-ones, through the zinc triflate-catalyzed Biginelli-type reaction of beta-keto-phosphonates, aldehydes and urea. The compounds obtained were characterized by various spectroscopic tools including IR, NMR (H-1, P-31, C-13) spectroscopy, mass spectrometry and single crystal X-ray diffraction. All the synthesized compounds were screened, for the first time, for anti-inflammatory activity by carrageenan-induced hind paw edema method, using female Wister rats and they showed significant anti-inflammatory activity in some cases higher than the standard indomethacin.
