Physical transfection technologies for macrophages and dendritic cells in immunotherapy

Summary: Dendritic cells (DCs) and macrophages, key antigen presenting cells of the innate immune system, are being explored for cell-based cancer immunotherapy by delivering functional macromolecules like mRNA and pDNA into their cytosol. Physical intracellular delivery techniques like electroporation, sonoporation, nanowires, and microfluidic cell squeezing are known to be more straightforward, controllable, faster, and efficient compared to viral and non-viral vectors. Further detailed studies on the impact of these delivery techniques on APC fitness, especially at the genetic level, are recommended to fully uncover their suitability for transfecting APCs with different types of macromolecules.
