This study demonstrates the screening of metal-binding pharmacophores against a zinc-dependent metalloenzyme using F-19 NMR, showing the potential of F-19 NMR-based techniques for ligand discovery in metalloenzymes.
This study demonstrates the screening of a collection of twelve F-19-tagged metal-binding pharmacophores (MBPs) against the Zn(ii)-dependent metalloenzyme human carbonic anhydrase II (hCAII) by F-19 NMR. The isomorphous replacement of Zn(ii) by Co(ii) in hCAII produces enhanced sensitivity and reveals the potential of F-19 NMR-based techniques for metalloenzyme ligand discovery.
