Development and Scale-Up of a Direct Asymmetric Reductive Amination with Ammonia

Summary: Direct asymmetric reductive amination is an efficient method for converting ketones to alpha-chiral primary amines, but reported examples are limited. This study developed two sets of Ru-catalyzed conditions for direct conversion of aryl methyl ketone to pharmaceutically relevant chiral primary amine, achieving >93% ee on multikilogram scale.

Summary: The asymmetric hydrogenation of enamines is efficiently catalyzed by a rhodium catalyst complexed with a fluorinated planar chiral paracyclophane-diphosphine ligand, showing high activity. This catalyst has also been successfully applied to Direct Asymmetric Reductive Amination, producing several tertiary amines with moderate enantioselectivity.

Chiral cyclometalated iridium complexes for asymmetric reduction reactions

Summary: A series of chiral cyclometalated iridium complexes were synthesized and studied for asymmetric transfer hydrogenation reactions using formic acid as the hydrogen source, showing different activities and enantioselectivities, with the most effective ones achieving up to 63% ee in asymmetric reductive amination of ketones and 77% ee in reduction of pyridinium ions.
