Going the dHis-tance: Site-Directed Cu2+ Labeling of Proteins and Nucleic Acids

Austin Gamble Jarvi et al.

Summary: This Account showcases the use of site-directed Cu2+ labeling in proteins and DNA for measuring biomolecular structure and dynamics with electron paramagnetic resonance (EPR) spectroscopy. The labeling scheme provides a straightforward method for obtaining biophysical information that is not accessible using traditional EPR labels. The integration of molecular dynamics (MD) simulations with EPR distance information is expected to advance our understanding of protein and DNA conformational changes and interactions.

Reimagining Druggability Using Chemoproteomic Platforms

Jessica N. Spradlin et al.

Summary: The majority of the proteome is currently considered undruggable due to the lack of well-defined binding pockets, requiring innovative approaches like chemoproteomic platforms. These platforms utilize chemical probes and quantitative mass spectrometry to discover ligandable sites and have revealed unique insights into ligandability, as well as expanding the scope of targeted protein degradation therapies through the discovery of new covalent E3 ligase recruiters. Exciting days lie ahead in the field of chemical biology as chemoproteomic approaches become a major driver in drug discovery.
