Revealing the Similarities of α,β-Unsaturated Iminiums and Acylazoliums in Organocatalysis

Summary: Secondary amine-catalyzed reactions via alpha,beta-unsaturated iminiums and N-heterocyclic carbene (NHC)-catalyzed transformations via alpha,beta-unsaturated acylazoliums are widely used electrophilic intermediates in organocatalysis for enantioselective construction of valuable molecules. Despite displaying similar reactivity patterns and conjugate additions of soft nucleophiles, there are subtle differences in reactivity between these intermediates. This Minireview aims to highlight the resemblances in reactivity between alpha,beta-unsaturated iminiums and acylazoliums in organocatalysis.

Summary: The enantioselective synthesis of 5,6-dihydroindolizines was achieved via N-heterocyclic carbene (NHC) catalysis, involving a cascade process with alpha,beta-unsaturated aldehydes and suitably substituted pyrrole derivatives, leading to efficient yields and high selectivities. The method was also extended to the construction of spirocyclic 5,6-dihydroindolizines.

Summary: A carbene-catalyzed addition reaction between isothioureas and enals under oxidative conditions has been reported, resulting in the formation of two carbon-nitrogen bonds in a highly regio-selective manner. Various sulphur-containing 5,6-dihydropyrimidin-4-ones have been obtained with high optical purity.
