Site-Directed Mutations of Calcium-Binding Sites Contribute to Reducing the Immunoreactivity of the EF-Hand Sarcoplasmic Calcium-Binding Protein in Scylla paramamosain

Summary: Site-directed mutations were utilized to reduce the immunoreactivity of sarcoplasmic calcium-binding protein (SCP) by replacing key amino acids in conformational epitopes and calcium-binding sites. The mutant SCPs effectively inhibited the binding with wild-type SCP and downregulated the expression levels of CD63 and CD203c on basophil surface, indicating impaired calcium binding ability and reduced immunoreactivities. This approach showed potential for application in reducing immunoreactivities of other shellfish allergens.

Mapping and IgE-binding capacity analysis of heat/digested stable epitopes of mud crab allergens

Summary: After high temperature-pressure treatment, the allergenic potential of tropomyosin and arginine kinase in mud crab was evaluated, showing higher prevalence and sensitivity to heat processing. New epitopes were identified among the 35 stable epitopes, and seven heat/digested stable peptides retained IgE-binding capacity.

Identification and Amino Acid Analysis of Allergenic Epitopes of a Novel Allergen Paramyosin (Rap v 2) from Rapana venosa

Summary: This study identified seven allergenic epitopes of the novel allergen Rap v 2 in sea snail, as well as important amino acid residues related to the formation of paramyosin epitopes. Three potential critical amino acids that may cause cross-reactivity between tropomyosin and paramyosin in molluscs were also discovered. These findings may have significant implications for understanding paramyosin epitopes and improving mollusc allergy prevention and therapy.

Effects of thermal processing on the allergenicity, structure, and critical epitope amino acids of crab tropomyosin

Summary: This study found that high temperature-pressure processing during food processing reduces the immunoreactivity of crab allergens by altering the protein structure. By identifying critical amino acids, the IgE binding activity of tropomyosin can be more accurately understood, helping food processing to reduce or eliminate the immunoreactivity of crabs.
