Cell therapy for advanced liver diseases: Repair or rebuild

Benjamin J. Dwyer et al.

Summary: Advanced liver disease poses a significant global health and economic burden, with liver transplantation as the only effective treatment option, but accompanied by risks and donor organ shortage. Cell therapies have advanced into early clinical trials, but challenges still need to be overcome before routine clinical use.

Protection of liver sinusoids by intravenous administration of human Muse cells in a rat extra-small partial liver transplantation model

Yoshihiro Shono et al.

Summary: Muse cells have a stronger ability to home to the liver and repair damaged tissue compared to MSCs, improving blood chemistry and proliferative hepatocytes within a short timeframe. Knockdown of hepatocyte growth factor and vascular endothelial growth factor-A can impair the protective effect of Muse cells on LSECs. Intravenous injection of Muse cells may be a feasible approach for LDLT with less recipient burden.

Downregulation of augmenter of liver regeneration impairs the therapeutic efficacy of liver epithelial progenitor cells against acute liver injury by enhancing mitochondrial fission

Yuan Dong et al.

Summary: Cell-based therapeutic approaches have shown to be effective in treating acute liver injury, but face limitations including stemness loss, aberrant differentiation, and low engraftment. Transplanting ALR-transfected LEPCs into ALI mice significantly protected against injury, reducing ALT and AST levels and inflammation, highlighting potential for clinical therapy.

Inhibition of Drp1 SUMOylation by ALR protects the liver from ischemia-reperfusion injury

Jing Huang et al.

Summary: This study demonstrated that ALR protects mitochondria by affecting Drp1 SUMOylation, which in turn rescues hepatocytes from IRI-induced apoptosis. The findings provide a potential target for clinical intervention to reduce the effects of IRI during hepatic surgery.
