Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Multiplexed and amplified chemiluminescence resonance energy transfer (CRET) detection of genes and microRNAs using dye-loaded hemin/G-quadruplex-modified UiO-66 metal-organic framework nanoparticles

Summary: In this study, dye-loaded UiO-66 metal-organic framework nanoparticles modified with catalytic hemin/G-quadruplex DNAzyme labels were utilized for chemiluminescence resonance energy transfer (CRET) analysis of miRNAs and genes. The engineered hairpin probes, caged G-quadruplex domains, and self-assembly of hemin/G-quadruplex DNAzyme labels allowed for sensitive detection of specific miRNAs and genes. The DNA hairpin-functionalized dye-loaded NMOF sensing modules were also applied for amplified miRNA or gene CRET-based sensing platforms, demonstrating multiplexed detection capabilities.

Snapshotting the transient conformations and tracing the multiple pathways of single peptide folding using a solid-state nanopore

The new single-molecule technique, combined with a Markov chain model, clarified non-equilibrium fluctuations in the transition pathways of the beta-hairpin peptide. This method provides high-throughput investigation of transition pathways, offering insights into protein folding mechanisms.

Single-molecule fluorescence detection of a tricyclic nucleoside analogue

Summary: A new tricyclic pyrimidine nucleoside analogue named ABN was designed and synthesized, which exhibited improved brightness and fluorescence properties in water, becoming the brightest and most red-shifted nucleobase analogue known in duplex DNA. Single-molecule fluorescence measurements of ABN showed excellent photostability with high count rates, offering robust and accessible single-molecule fluorescence detection capabilities.
