Modulation of the Canonical Wnt Signaling Pathway by Dietary Polyphenols, an Opportunity for Colorectal Cancer Chemoprevention and Treatment

Hernan Villota et al.

Summary: In recent years, dietary polyphenols have garnered interest as potentially anti-carcinogenic compounds by modulating the Wnt/beta-Catenin signaling pathway to reduce the risk of colorectal cancer. Promising results from clinical trials suggest the potential implementation of these dietary polyphenols into preventative and therapeutic therapies for CRC.

Pharmacological blockage of transforming growth factor-β signalling by a Traf2-and Nck-interacting kinase inhibitor, NCB-0846

Teppei Sugano et al.

Summary: The small-molecule TNIK inhibitor NCB-0846 has been shown to inhibit TGF-beta signaling and nuclear translocation, thereby preventing EMT in lung cancer cells and reducing metastasis through transcriptional downregulation of TGFBRI expression. This represents a potentially promising approach for the treatment of lung cancer patients.

Hypoxia-Induced Suppression of Alternative Splicing of MBD2 Promotes Breast Cancer Metastasis via Activation of FZD1

Zhaoji Liu et al.

Summary: This study reveals that under hypoxic conditions, the alternative splicing of MBD2 is suppressed, leading to the production of MBD2a which promotes breast cancer metastasis. Clinical data show a significant correlation between the expression of MBD2a and MBD2c with the invasiveness of breast cancer, indicating their opposing roles in regulating metastasis in this disease.

Manipulating the Wnt/β-catenin signaling pathway to promote anti-tumor immune infiltration into the TME to sensitize ovarian cancer to ICB therapy

J. A. Wall et al.

Summary: This study evaluated the anti-tumor and immune-enhancing properties of the Wnt inhibitor CGX-1321 in ovarian cancer. Results showed that CGX-1321 significantly reduced tumor burden and increased CD8+ T cell infiltration. Although the addition of DKN-01 or anti-PD-1 therapies did not enhance the effects of CGX-1321, further investigation is needed to determine if CGX-1321 + DKN-01 combination treatment sensitizes pre-clinical ovarian cancer to ICB therapy.

Mutations and mechanisms of WNT pathway tumour suppressors in cancer

Jeroen M. Bugter et al.

Summary: Activation of WNT-beta-catenin signalling via mutations is a recurrent driver event in human cancer, granting cancer cells sustained growth properties and therapy resistance. Researchers reviewed the recent advances and challenges in understanding how different mutational subsets of WNT tumor suppressor genes link to distinct cancer types, clinical outcomes, and treatment strategies.
