Molecular Recognition and Imaging of Human Telomeric G-Quadruplex DNA in Live Cells: A Systematic Advancement of Thiazole Orange Scaffold To Enhance Binding Specificity and Inhibition of Gene Expression

Summary: The series of fluorescent ligands constructed from thiazole orange scaffold were investigated for their interactions with G-quadruplex structures and antitumor activity. Compound 3 showed excellent specificity towards telomere G4-DNA, with higher affinity compared to other nucleic acids. The findings support that compound 3 may be telomere G4-DNA specific and offer insights for anticancer drug design.

The G-quadruplex stabilization of a library of nine compounds was examined, identifying several compounds with interesting selectivity profiles. Compound 3, based on a perylene scaffold, was found to be the best G4 ligand with 40-fold selectivity.
