Folate Receptor Beta Designates Immunosuppressive Tumor-Associated Myeloid Cells That Can Be Reprogrammed with Folate-Targeted Drugs

Gregory M. Cresswell et al.

Summary: The immunosuppressive environment within tumors limits the efficacy of immunotherapies, with MDSCs and TAMs playing a key role in hindering immune cell functions. Targeting the FRβ(+) subset of these cells with folate-linked drugs can reprogram them to enhance antitumorigenic responses, inhibit tumor growth, block metastasis, and improve overall survival without toxicity. This strategy has broad applications for the treatment of various tumors.

Entinostat plus Pembrolizumab in Patients with Metastatic NSCLC Previously Treated with Anti-PD-(L)1 Therapy

Matthew D. Hellmann et al.

Summary: In patients with NSCLC who had previous experience with PD-(L)1, the combination of entinostat and pembrolizumab did not meet the primary response rate endpoint but provided a clinically meaningful benefit, with 9% of patients showing objective response. No new toxicities, including immune-related adverse events, were observed.

The multifaceted role of TRAIL signaling in cancer and immunity

Ludmila Cardoso Alves et al.

Summary: This review article examines the current knowledge on noncanonical TRAIL signaling, presenting and explaining the biological outcomes of TRAIL signaling in immune and malignant cells with a focus on its role in NK cell function. The challenges in dissecting the precise molecular mechanisms involved in TRAIL signaling and the implications for therapeutic manipulation in cancer are discussed, along with possible approaches to overcome these difficulties.

Effect of Taxane Chemotherapy With or Without Indoximod in Metastatic Breast Cancer A Randomized Clinical Trial

Veronica Mariotti et al.

Summary: In a prospective clinical trial, the addition of the IDO1 pathway inhibitor indoximod to a taxane did not improve progression-free survival in patients with ERBB2-negative metastatic breast cancer compared to a taxane alone. This study highlights the importance of exploring new treatment strategies for overcoming tumor immune suppression in breast cancer.

The Functional Crosstalk between Myeloid-Derived Suppressor Cells and Regulatory T Cells within the Immunosuppressive Tumor Microenvironment

Maximilian Haist et al.

Summary: Immunotherapy has shown benefits for advanced cancer patients, but many still do not respond to treatment. The accumulation of immunosuppressive cell populations like MDSC and Treg in the tumor microenvironment contributes to immune resistance and limits the effectiveness of immunotherapy.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

MDSC: Markers, development, states, and unaddressed complexity

Samarth Hegde et al.

Summary: MDSCs are a widely discussed biological entity in immunology, commonly describing cells that arise during chronic inflammation with T cell immunosuppressive functions. Currently lacking clear definitions and a unifying framework across pathologies, we propose a framework based on activation signals to classify MDSCs as discrete cell states. Developing this knowledge of myeloid states across pathological conditions may transform how diseases are grouped and treated.

Blocking IL1 Beta Promotes Tumor Regression and Remodeling of the Myeloid Compartment in a Renal Cell Carcinoma Model: Multidimensional Analyses

David H. Aggen et al.

Summary: The study suggests that intratumoral immunosuppression mediated by myeloid-derived suppressor cells and tumor-associated macrophages may lead to immune checkpoint inhibitor resistance in solid tumors. Blocking IL1 beta in combination with anti-PD-1 or cabozantinib has shown increased antitumor activity by reducing immunosuppressive myeloid cells and increasing M1-like tumor-associated macrophages.

Summary: MDSCs are pathologically activated cells with potent immunosuppressive activity, closely associated with poor clinical outcomes in diseases like cancer. Recent studies have identified key distinctions between MDSCs and classical neutrophils and monocytes, providing new insights into therapeutic targeting for cancer and autoimmune diseases through understanding their genomic and metabolic characteristics. Emerging data also suggest potential involvement of MDSCs in pregnancy, neonatal biology, and COVID-19.

Retinoic Acid Synthesis Deficiency Fosters the Generation of Polymorphonuclear Myeloid-Derived Suppressor Cells in Colorectal Cancer

Hong-Wei Sun et al.

Summary: Metabolism in cancer is reprogrammed to meet the demands of malignant cells, and the deficiency in retinoic acid synthesis may contribute to the generation of PMN-MDSCs in colorectal cancer. Restoring RA signaling in the TME could be a promising therapeutic strategy to inhibit the generation of PMN-MDSCs.
