Non-negative assisted principal component analysis: A novel method of data analysis for raman spectroscopy

The method allows derivation of physically realistic spectra and analysis of chemical and spatial trends across a sample surface. Multivariate methods outperform univariate analysis for the analysis of hyperspectral SRS data. Among the methods tested, multivariate curve resolution (MCR) gives the best results, showing a simple workflow for the analysis of hyperspectral coherent Raman imaging data.

Computational techniques are applied for spectral decomposition and feature selection to identify a sparse subset of Raman frequencies capable of sample discrimination.

Large datasets of chemical images can be obtained in a reasonable timescale, leading to improved biochemical modeling and identification of rare cells.
