Chiral Cyclopentadienyl Ligands: Design, Syntheses, and Applications in Asymmetric Catalysis

Josep Mas-Rosello et al.

Summary: The development of new chiral ligands is crucial for stereocontrol in metal-catalyzed reactions, especially with the increasing demand for bioactive molecules as single enantiomers. The successful application of Cp-X ligands in metal-catalyzed transformations allows access to valuable chiral molecules, with critical comparisons of existing ligand families and discussion of future research directions for further enhancing their performance and application in enantioselective catalysis.

Air Stable Iridium Catalysts for Direct Reductive Amination of Ketones

Iuliia Polishchuk et al.

Summary: Half-sandwich iridium complexes with bidentate urea-phosphorus ligands were found to catalyze the direct reductive amination of ketones with high selectivity under mild conditions. One of the complexes showed activity in two different reactions and the use of ammonium formate does not require special conditions, taking place in HFIP instead of methanol.

Iridium-Catalyzed Acid-Assisted Hydrogenation of Oximes to Hydroxylamines

Josep Mas-Rosello et al.

Summary: Cyclometalated cyclopentadienyl iridium(III) complexes are efficient catalysts in the hydrogenation of oximes, with alkoxy-substituted aryl ketimine ligands providing the best catalytic performance. The stable iridium C,N-chelation is crucial for the catalytic activity, and the hydrogenation system demonstrates high functional group compatibility.

Highly Enantioselective Direct Asymmetric Reductive Amination of 2-Acetyl-6-Substituted Pyridines

Masatoshi Yamada et al.

Summary: This study presents a highly efficient direct asymmetric reductive amination, successfully achieving reactions on various ketone substrates for the first time, including 2-acetyl-6-substituted pyridines. Excellent enantioselectivity and conversion rates were achieved through the use of commercially available and inexpensive Ru(OAc)(2){(S)-binap} catalyst under 0.8 MPa of hydrogen gas pressure.

Recent advances on transition-metal-catalysed asymmetric reductive amination

Yingying Tian et al.

Summary: Transition-metal-catalysed asymmetric reductive amination is a direct method for accessing chiral amines, converting carbonyl compounds and ammonia or amines into drug-relevant compounds in one pot. Recent progress has been made in homogeneous transition-metal-catalysed ARA reactions with H-2 or other hydride sources, but challenges and potential still exist in this field.

Chiral cyclometalated iridium complexes for asymmetric reduction reactions

Jennifer Smith et al.

Summary: A series of chiral cyclometalated iridium complexes were synthesized and studied for asymmetric transfer hydrogenation reactions using formic acid as the hydrogen source, showing different activities and enantioselectivities, with the most effective ones achieving up to 63% ee in asymmetric reductive amination of ketones and 77% ee in reduction of pyridinium ions.
