Synthetic Biological Circuits within an Orthogonal Central Dogma

Alan Costello et al.

Summary: Researchers aim to control cellular behavior by reducing or eliminating unwanted interactions, in order to achieve reliable and predictable gene circuits and develop context-independent biological activities. Current focus is on biological orthogonalization, isolating researcher-specified bioactivities from host processes, with the potential to create an orthogonal central dogma and novel cellular functions.

Engineering a Highly Efficient Carboligase for Synthetic One-Carbon Metabolism

Maren Nattermann et al.

Summary: This study successfully converted OXC into GCS through enzyme mutation, enabling the direct condensation of two C1 units with high catalytic efficiency. The new variant MeOXC4 showed a catalytic efficiency in C1-C1 condensations more than 40 times higher than other enzymes, and was able to produce glycolate in vivo, offering new possibilities for synthetic C1 assimilation routes.
