On-resin multicomponent protocols for biopolymer assembly and derivatization

Daniel G. Rivera et al.

Summary: Solid-phase synthesis and multicomponent reactions serve different purposes, with the former mainly used for the automated synthesis of large molecules and the latter for generating diversity in skeletal and appendage structures. While on-resin MCRs were initially employed for library construction, the current focus has shifted towards the derivatization of peptides and oligonucleotides.

Nickel-Catalyzed, Stereospecific C-C and C-B Cross-Couplings via C-N and C-O Bond Activation

Jianyu Xu et al.

Summary: The development of mild, nickel-catalyzed cross-couplings for the preparation of highly enantioenriched products, including those with quaternary stereocenters, has expanded the synthetic toolbox. This perspective highlights the use of convenient and functional-group-tolerant organoboron reagents, as well as the discoveries of activating groups and conditions that have led to highly stereospecific reactions. Emphasis is placed on mechanistic understanding and controlling inversion vs retention pathways, while also discussing limitations and future development opportunities.

Mechanism-Guided Development of Transition-Metal-Catalyzed C-N Bond-Forming Reactions Using Dioxazolones as the Versatile Amidating Source

Seung Youn Hong et al.

Summary: Catalytic reactions constructing carbon-nitrogen bonds are important in synthetic and medicinal chemistry due to the prevalence of nitrogen-containing motifs in natural products and drugs. C-H amination offers a more efficient approach by directly converting C-H bonds into C-N bonds, bypassing the need for prefunctionalized substrates. The development of novel catalytic platforms utilizing metal-nitrenoids has expanded the scope of C-N bond-forming reactions and enabled the synthesis of diverse compounds.

1,2,3-Triazoles as Biomimetics in Peptide Science

Naima Agouram et al.

Summary: The potential use of peptides as drugs is limited by their low bioavailability and enzymatic degradation in vivo, and the development of new peptide-mimicking molecules is crucial. Replacing the amide bond in peptides with a heterocyclic bioisostere, such as the 1,2,3-triazole ring, can lead to the synthesis of biologically active peptidomimetics with interesting biological activities. Triazoles can also serve as linker moieties to connect peptides to other functional groups, making them versatile tools in drug development.

Organoboron compounds as versatile reagents in the transition metal-catalyzed C-S, C-Se and C-Te bond formation

Angelita M. Barcellos et al.

Summary: Organochalcogen compounds have significant synthetic and biological applications. The coupling reaction between boronic acids, borate anions or boronic esters with different sources of chalcogen mediated by transition metals is a powerful tool for the formation of functionalized diorganyl chalcogenides. This strategy allows for the preparation of selenides, sulfides, and tellurides with high complexity and molecular diversity in good to excellent yields.

Nickel-catalyzed C-O/N-H, C-S/N-H, and C-CN/N-H annulation of aromatic amides with alkynes: C-O, C-S, and C-CN activation

Yasuaki Iyori et al.

Summary: The Ni-catalyzed reaction involves annulation of ortho-phenoxy-substituted aromatic amides with alkynes to form 1(2H)-isoquinolinones in the presence of (LiOBu)-Bu-t as a base. The reaction proceeds even without a ligand under mild conditions at 40 degrees C. An electron-donating group on the aromatic ring facilitates the reaction, which is also applicable to various leaving groups such as carbamate, methylthio, and cyano.
