Final Efficacy Results of Neratinib in HER2-positive Hormone Receptor-positive Early-stage Breast Cancer From the Phase III ExteNET Trial

Summary: In early-stage breast cancer patients, neratinib provides a 5.1% increase in invasive disease-free survival and a 2.1% increase in overall survival at 8 years compared to placebo. For patients with residual disease after neoadjuvant therapy, the absolute benefits of neratinib are 7.4% and 9.1%, respectively.

Novel oxindole/benzofuran hybrids as potential dual CDK2/GSK-3β inhibitors targeting breast cancer: design, synthesis, biological evaluation, and in silico studies

Summary: In this study, three series of oxindole-benzofuran hybrids were designed and synthesised as dual CDK2/GSK-3 beta inhibitors targeting breast cancer. The N-1 -unsubstituted oxindole derivatives showed potent cytotoxic activity, while the N-1-substituted derivatives exhibited moderate to weak activity on breast cancer cell lines. The most potent compounds caused cell cycle arrest in the G2/M phase by inhibiting CDK2/GSK-3 beta, as demonstrated by enzyme inhibition assays and molecular docking studies.

Summary: The study identified a redox homeostasis network involving the iron-sulfur cluster enzyme NFS1 in the survival mechanisms governed by the tumor hypoxia-induced pH regulator CAIX. Targeting CAIX while depleting NFS1 or blocking cyst(e)ine availability enhanced ferroptosis and inhibited tumor growth significantly. Altering intracellular pH and iron homeostasis through inhibiting CAIX activity may lead to the development of innovative therapeutic strategies for solid tumors to overcome hypoxia- and acidosis-mediated tumor progression and therapeutic resistance.
