Biocatalysis: Enzymatic Synthesis for Industrial Applications

Carboxylic Acid Reductase Can Catalyze Ester Synthesis in Aqueous Environments

Summary: Most esterification enzymes require minimization of water or activated substrates for activity, but this study found that carboxylic acid reductase (CAR) can catalyze esterification under aqueous conditions. By adding imidazole and other co-substrates, CAR can achieve high esterification yields for the synthesis of various compounds.

Summary: A highly atom-efficient integrated cofactor/co-product recycling cascade using cycloalkylamines for the synthesis of nylon building blocks was reported, achieving excellent conversions with E. coli whole cells and purified enzymes. The tandem biocatalytic cascade was demonstrated to produce lactams using engineered biocatalysts, showing promise as an alternative for bio-based production of omega-amino acids and corresponding lactam compounds.

Whole-cell biocatalysis using the Acidovorax sp. CHX100 Δ6HX for the production of ω-hydroxycarboxylic acids from cycloalkanes

Summary: This study investigates the use of an Acidovorax mutant as a whole-cell biocatalyst for converting cycloalkanes to omega-hydroxycarboxylic acids, showcasing high metabolic efficiency with an efficient enzyme cascade. The deletion of a specific enzyme in the metabolic pathway led to the production of short-chain omega-hydroxycarboxylic acids, demonstrating a high molar conversion rate above 98% within 6 hours.

Summary: A bienzymatic cascade was developed for the synthesis of beta-amino acids as an intermediate for sitagliptin production. Recombinant Escherichia coli were used for whole-cell biotransformation to achieve high conversion rates. The desired product was obtained with excellent conversions in small-scale reactions, and sitagliptin phosphate was synthesized with high yield and purity.

To overcome this, researchers have been devoted to expanding the protein sequence space through chemical modifications and/or incorporation of noncanonical amino acids.
