Summary: Tuberous sclerosis complex is caused by the loss of tumor suppressor genes TSC1 or TSC2, which encodes hamartin and tuberin respectively. Gene therapy using adeno-associated virus vectors carrying a condensed form of human tuberin has shown potential in treating life-threatening TSC2 lesions. The survival of a mouse model with TSC2 was extended and brain pathology reduced after a single intravenous injection of AAV9-cTuberin.

The novel STRADA missense mutation c.792T>A (p.Ser264Arg) in exon 10 was identified in a female patient with typical clinical features of PMSE, contributing to a better understanding of this syndrome.

Clinical experience with the AKT1 inhibitor miransertib in two children with PIK3CA-related overgrowth syndrome

Treatment was well tolerated with no significant toxicities reported, highlighting the potential therapeutic utility of miransertib in selected pediatric patients with severe PROS and demonstrating the possibility of repurposing targeted therapies for rare diseases. An open label, Phase 1/2 study of miransertib in children with PROS and PS is currently underway to more accurately assess its efficacy in treating PROS disorder.

The study demonstrates that cell-free DNA in cerebrospinal fluid is brain-derived and may aid in investigating mosaic neurological disorders where brain tissue is unavailable.
