ADO09, a co-formulation of the amylin analogue pramlintide and the insulin analogue A21G, lowers postprandial blood glucose versus insulin lispro in type 1 diabetes

Grit Andersen et al.

Summary: This study compared the safety, pharmacokinetics, and pharmacodynamics of ADO09 with insulin lispro and separate subcutaneous injections of human insulin and pramlintide in subjects with type 1 diabetes. Results showed that ADO09 significantly reduced postprandial blood glucose excursions compared to insulin lispro and displayed similar gastric emptying kinetics to Ins&Pram. The positive results support further investigations with ADO09.

Lentivirus Mediated Pancreatic Beta-Cell-Specific Insulin Gene Therapy for STZ-Induced Diabetes

Fulya Erendor et al.

Summary: Autoimmune destruction of pancreatic beta cells is the hallmark of type 1 diabetes, prompting the development of insulin analogs to compensate for the deficiency. Basal and post-prandial insulin secretion, as well as gene therapy, are being explored as potential treatments. The lentiviral vector LentiINS has shown promising results in lowering blood glucose and improving tolerance in diabetic rats, but combinatorial use with anti-inflammatory lentiviral vector LentiVIP is necessary for more significant effects.

Summary: Insulin is commonly delivered through subcutaneous injections for the treatment of diabetes, which can be invasive and hinder patient compliance. Transdermal delivery methods, such as microneedles and patches, offer advantages including prolonged drug release and improved patient adherence. Research in transdermal insulin delivery is ongoing, and it is expected that these systems may soon be introduced in clinical practice.

Switching to Once-Weekly Insulin Icodec Versus Once-Daily Insulin Glargine U100 in Type 2 Diabetes Inadequately Controlled on Daily Basal Insulin: A Phase 2 Randomized Controlled Trial

Harpreet S. Bajaj et al.

Summary: The study demonstrated that switching from daily basal insulin to once-weekly icodec was well tolerated and provided effective glycemic control. The use of a loading dose when switching to once-weekly icodec significantly increased the percent time in range during weeks 15 and 16 compared to once-daily IGlar U100, without increasing the risk of hypoglycemia.
